If there is one slide that consistently overwhelms investors, it’s the mechanism of action slide.

Founders often compress entire pathways into a single diagram: every receptor, ligand, arrow, cell type, and downstream effect piled into a dense scientific maze. It’s accurate, certainly. But accuracy is not the same as communication. Investors stop following the talk track, the story stalls, and a program that may be compelling loses its momentum simply because the biology is hard to parse.

The deeper issue is not complexity. It’s missing context. Many teams jump straight to “what our therapy does” without showing the two things that make the mechanism meaningful in the first place: how the system operates in its normal physiological state and where pathway dysfunction or loss of regulatory control emerges in disease. Without that contrast, the therapeutic action feels disconnected, almost abstract, and investors are left struggling to assemble cause and effect on their own.


 

Why most MOA slides fail

Mechanisms fail to resonate with investors for two predictable reasons.

First, companies try to communicate everything at once. They place the entire molecular sequence on a single canvas and attempt to describe it verbally in one breath. The viewer’s attention fragments. Without hierarchy, every arrow seems equally important, and the essential insight disappears.

Second, teams often skip the healthy baseline. Scientists know what “normal” looks like, so they leap directly into the malfunctioning state. But an investor who hasn’t spent a decade immersed in the pathway has no reference point. They can’t see what’s wrong because they never saw what “right” looked like. Without a baseline, the disease panel becomes noise.

If the baseline is missing and the diagram is overloaded, the mechanism stops being a mechanism and becomes a puzzle.

 

Your MOA should read like a short story

A compelling MOA explanation mirrors the logic of a simple narrative: establish normalcy, introduce the disruption, and then reveal the solution. Each part depends on the previous one, and clarity increases as the story unfolds.

In practice, this translates into three consecutive panels:

  1. Healthy state
  2. Disease state
  3. Therapeutic action

This sequence is not cosmetic. Investors think in causal terms. They want to understand what is broken in the biology and how your program repairs it. When you give them this progression, they can follow your reasoning with ease.

Let’s break down each component.

1) Healthy state: anchor the viewer

Start by showing the pathway or cellular process in its simplest, fully functional form. The goal is not to provide an encyclopedic illustration but to establish how the system behaves when everything is in balance. You are giving the audience a reference frame.

Describe only the essential actors and show the basic sequence of events. When investors see the purpose of the pathway, whether it’s enabling immune activation, maintaining homeostasis, or supporting cell survival, they understand why the upcoming disruption is meaningful. The healthy panel should be so clear that the viewer can explain it back in one sentence.

This panel should answer a single question: how does this system work when nothing is wrong? When that answer is clear, scientific complexity becomes intuitive understanding.

2) Disease state: show what breaks

Once the baseline is established, guide the viewer to the disease state by highlighting the precise point where the biology falls apart. The most effective disease panels maintain the same layout and core visual elements as the healthy panel, allowing investors to instantly spot what has changed. Consistency here dramatically improves comprehension.

Instead of listing every consequence of the dysfunction, focus on the biological event that drives pathology. Is a receptor mutated and unable to signal? Does a ligand fail to bind? Does a step in the cascade become overactive or insufficient? Clarify the disruption and briefly describe how it leads to the observed effect.

This section should answer a single question: what exactly is the problem your therapy must solve?

When done well, investors can grasp the disease logic without effort, and the therapeutic action that follows feels like a natural next step.

3) Therapeutic action: reveal how the drug fixes the problem

With the problem clearly defined, you can now show how your therapeutic agent intervenes. This is the moment that connects the scientific rationale to the clinical promise.

Introduce the drug into the same diagram and show how it restores function, compensates for loss, or counteracts hyperactivity. Describe its role using one strong, active verb and trace the downstream effect only so far as needed to show biological rescue. Investors do not need every branch of the pathway. They need the segment that proves your drug’s impact is meaningful, measurable, and linked to the clinical outcomes you will present later.

This panel should answer one question: does your drug create a change in biology that is directly linked to what you will measure in the clinic?

When the three panels read as one coherent sequence, investors leave with a clear understanding of how and why your therapy works.

Your MOA slides often go wrong

Even with the right structure, several recurring mistakes undermine clarity.

Overloading the therapeutic panel with secondary biologyWhen teams add too many downstream effects into the solution panel, it stops reading as a resolution and starts feeling like a second problem statement. Show only the segement of the pathway that proves your drug’s impact is meaningful and directly linked to clinical outcomes.

Mixing mechanism and data on the same slide – Placing preliminary results or efficacy signals alongside the MOA forces investors to interpret two different narratives at once. And when multiple mechanisms are presented in a single diagram, the viewer is left unsure which action actually matters. Data belongs on a separate slide, where it can be presented with the context it deserves.

Designing complex MOA diagrams with multiple mechanisms or pathways If your program has multiple mechanisms, prioritize the one that carries the most clinical weight and address others separately. A visually polished illustration that lacks logical flow confuses more than a simple diagram that guides the viewer from cause to effect. The strongest MOA slides prioritize coherence: clean visuals, minimal clutter, and a sequence that clearly answers what’s broken and how the therapy repairs it.


 

A Clear MOA strengthens the entire investor presentation

A well-crafted MOA illustration is more than a scientific explainer. It is a central component of an investor presentation, helping investors understand the scientific logic behind your program. It clarifies why you chose your target, sets up your pre-clinical and clinical data, and reinforces the core value proposition of your therapeutic approach. When investors understand the mechanism, everything that follows in the deck feels more credible.
